摘要
目的 观察 L -苏糖酸盐对体外兔破骨细胞 (OC)骨吸收功能的影响。方法 制备骨磨片 ,培养 OC,并分别加入高中低浓度 (10 - 5、10 - 7、10 - 9m ol/L )的 L -苏糖酸钙、L -苏糖酸钠、阿仑膦酸钠、17β-雌二醇、葡萄糖酸钙 ,实验设以上各浓度组及对照组 (共 16组 ,每组 n=8) ,甲苯胺蓝染色 -光镜观察分析骨片上骨吸收陷窝面积 ,EL ISA测定上清液 型胶原 C-末端肽 (CTx或 Crosslaps)浓度。结果 1L -苏糖酸钙各组陷窝面积及 CTx浓度均较葡萄糖酸钙组及对照组减少 (P<0 .0 0 1) ;L -苏糖酸钠各组作用较对照组显著 (P<0 .0 5 ) ;而葡萄糖酸钙组与对照组无显著差异 (P>0 .0 5 )。 2同浓度 L -苏糖酸钙较 L -苏糖酸钠组陷窝面积、CTx浓度减少显著 (P<0 .0 5 ) ;按 CTx水平 ,低、中浓度 L -苏糖酸钙组达到或超过中、高浓度 L -苏糖酸钠组作用 (P<0 .0 5 )。 3高浓度 L -苏糖酸钙组作用介于中、低浓度 17β-雌二醇组之间 (P<0 .0 5 ) ,但无阿仑膦酸钠各组显著 (P<0 .0 0 1) ;4上清液 CTx浓度与陷窝面积高度相关 (r=0 .876 )。 5 CTx在多个组间比较时显示比陷窝面积更敏感、更稳定 (平均变异系数分别为 10 .0 1%vs14 .6 6 %)。结论 L -苏糖酸盐尤其是 L -苏糖酸钙在体外有一定的抑制破骨细胞骨吸收的作用 。
Objective To clarify if calcium L-threonate and sodium L-threonate have inhibitory effects on the bone resorption of rabbit's osteoclasts in vitro. Methods This study contained a total of 16 culture groups, including one group as control and 5 groups treated by 5 drugs(calcium L-threonate, sodium L-threonate, alendronate, 17β-estradiol and calcium gluconate) each at the final concentrations of 10 -9 mol/L, 10 -7 mol/L, 10 -5 mol/L respectively. After 7 days, eight bone slices of every group were stained with toluidine blue and the areas of resorptive pits were analyzed under light microscope; the concentrations of C-telopeptide of typeⅠcollagen (CT X or Crosslaps) in culture supernatants were measured by ELISA. Results ①The resorption area and the CT X concentration of the Calcium L-threonate groups were reduced significantly as compared with those of control and of Calcium gluconate groups respectively. The resorption area and CT X level of the Sodium L-threonate groups were significantly reduced when compared with those of the control, but the effects of Calcium gluconate groups were not so. ② The reduction in the resorption area and CT X concentration of Calcium L-threonate group was more than that of Sodium L-threonate group. ③ The reductive effect of the high concentration(10 -5 ) group of Calcium L-threonate on the area and CT X level was corresponding to that of 17β-estradiol at a concentration between 10 -7 and 10 -9 . ④ The resorption area was related to the CT X concentration(r=0.876). ⑤ The CTX level was much more sensitive, precise and stable than the concentration. Conclusion L-threonate, especially calcium L-threonate could inhibit the bone resorption of osteoclasts in vitro, and its effect might be related to the radical of L-threonic acid. The CT X concentration in culture supernatants might be an effective marker quantitatively reflecting the bone resorption by osteoclasts in vitro.
