肺腺癌细胞缺氧后移植瘤生长特性及微血管密度变化

摘要 目的探讨肺腺癌细胞经过缺氧刺激后,对肿瘤生长及血管形成的影响。方法将人肺腺癌细胞株A549暴露于常氧(空气,5%CO2)、缺氧(5%O2,5%CO2,90%N2)、无氧(95%N2,5%CO2)环境48h后,将细胞接种于裸鼠皮下,观察其生长情况,通过CD34免疫组化染色计数微血管密度(MLD),通过ELISA测定移植瘤组织中血管内皮细胞生长因子(VEGF)及碱性成纤维细胞生长因子(bFGF)的表达水平。结果移植10d之后,缺氧组肿瘤体积显著大于常氧组。移植25d之后缺氧组肿瘤体积、重量、微血管密度以及瘤组织中VEGF、bFGF水平均显著高于常氧组,而无氧组相反,显著低于常氧组。结论适度缺氧可以刺激肺腺癌VEGF、bFGF等生长因子的表达,促进移植瘤血管形成,使肿瘤的生长能力提高。但是严重缺氧可损伤癌细胞,影响生长因子的合成表达,阻碍血管形成,抑制肿瘤的生长。 Objective To realize the effect of hypoxia on the growth and angiogenesis non-small cell lung carcinoma. Methods Non-small cell Lung carc inoma cell (A549) were exposed to hypoxia(5%O2,5%CO2,90%N2) or anoxia(95%N2, 5%C O2) condition for 48 hours. The cells exposed to hypoxia were planted under skin in nude mice, and observed the volume per 5 days. After 25 days, the weight and volume were measured, and the mirovessel density (MLD) were observed by immune tissue chemistry method for CD34, the level of vascular endothelial growth facto r (VEGF) and basic fibroblast growth factor (bFGF) were assayed by ELISA. Resu lts Comparing with normoxia group, the volume and weighty of tumor under skin of nude mice invasiveness were increased in hypoxic group, the MLD and the leve l of VEGF and bFGF in hypoxic group were higher than those in normoxia group. Th e volume, the weighty, MLD and the level of VEGF and bFGF in anoxic groups, were decreased contrast to normoxic groups. Conclusions Moderate hypoxia can up- regulated the expression of VEGF and bFGF, and increase the angiogenesis and gro wth of non-small cell lung carcinoma. But serious hypoxia can decrease the expre ssion of VEGF and bFGF, and inhibite the angiogenesis and growth of the carcinom a.
