摘要
研究目的探讨胺碘酮对肺的毒性作用及发生机制。研究背景目前胺碘酮肺损害的机制不清。由于各种原因临床研究受到限制,而动物研究甚少。方法通过气管内注入胺碘酮建立肺损害的豚鼠模型,将动物分为给药组与对照组,观察其组织病理学及超微结构变化同时测定血循环免疫复合物结果给药组全部出现不同程度的组织学异常并且与人类胺碘酮肺损害非常相似。给药组循环免疫复合物明显增高(P<0.05)。结论经气管内注入胺碘酮可在较短时间内制成理想的肺损害模型。肺损害的机制可能为细胞毒性及免疫异常的共同结果。
Objective To search the viruience action and mechanism of amiodarone on pulmonary.background At the present, the mechanism is not clear on amiodarone pulmonary damage. The clinical research is limtted by various factors, and the research in animais is few.Method A guinea pig model of puimonary damage is established through amiodarone poured into trachea. The animals were divided into two groupsf drug-group and control-group. Then the histopathology and ultrastructure were obsearved,and the circulating immune compfex (CIC) was measured at the same time.Result The histological abnormal was observed in all the drug - groups with varying degrees. This model appeared to be very similar to human's pulmonary damage induced by amiodarone The CIC are significantly raised in the drug- groups (P<0.05).Conclusion Satisfactory pulmonary damage model is established in short time through amiodarone poured into trachea. The study suggests the mechanism of virulence of amiodarone may be common results of cell toxicity and abnormal immunity.
