乳腺癌端粒酶CD_(44v)基因蛋白表达及临床意义 被引量:2

摘要 目的探讨端粒酶以及CD44v(细胞表面跨膜糖蛋白 )在乳腺癌发生、侵袭和转移中的作用 ,为临床治疗、判断预后提供依据。方法(1)实验组为50例原发性乳腺癌存档蜡块标本 ,对照组为5例正常乳腺组织。应用流式细胞术定量检测端粒酶和CD44v 基因蛋白表达。(2)统计学处理采用t检验、方差分析、q检验、χ2 检验和相关分析 ,显著性水准为0.05。结果(1)CD44v 在正常乳腺组织中表达阴性 ,在乳腺癌组织中表达阳性率为58 %(29/50) ,两组比较有显著性差异(P<0. 05)。(2)端粒酶在正常乳腺组织中表达阴性 ,在乳腺癌组织中表达阳性率为70 %(35/50) ,两组比较有显著性差异(P<0. 05)。(3)端粒酶与CD44v 表达之间呈正相关 (r=0.3333 ,P<0 .05 )。结论结果表明端粒酶、CD44v 基因蛋白均参与了乳腺癌的发生发展过程。它们以不同的作用机理促进了乳腺癌的发生、侵袭和转移。 Objectives To investigate the role of telomerase,CD 44v in carcinogenesis,invasion and metastasis of breast carcinoma to provide theoretical basis for clinical therapy and evaluating prognoˉsis.Methods(1)The experimental group contained50paraffin-embeded samples of breast Carciˉnoma.The control group contained5normal breast tissues.The expressions of Telomerase,CD 44v protein were detected by flow cytometry.(2)Significance was set at P<0.05.Students't-test,analˉysis of variance,q-test,X 2 -test,correlation analysis were carried out for statistic evaluation.Results(1)CD 44v gene protein expression was all negative in5normal breast tissues and its positive expression rate was58%(29/50)in50breast carcinoma.Comparision between two groups showed significant difference(P<0.05).(2)Telomerse expression was all negative in5normal breast tissues and its posiˉtive expression rate was70%(35/50)in50breast Carcinoma.Comparision between two groups showed significant difference(P<0.05).(3)Telomerse expression correlated positively with CD 44v protein expression(r=0.3333,P<0.05).Conclusions The results suggest that two proteins of telo_ merase and CD 44v were involved in the process of carcinogenesis and development of breast carcinoma.They contributed to the carcinogenesis,invasion and metastasis of breast carcinoma according to their different effects.
