低剂量辐射对荷人小细胞肺癌(NCI-H446)裸小鼠移植瘤细胞凋亡的影响 被引量:2

摘要 目的探讨低剂量辐射对荷人小细胞肺癌(NCIH446)裸小鼠移植瘤细胞凋亡的影响。方法对荷人小细胞肺癌裸小鼠进行全身深部X射线照射,采用TUNEL(末端脱氧核甘酸转移酶介导的dUTP缺口末端标记)检测荷人小细胞肺癌裸小鼠移植瘤组织细胞凋亡的情况。结果D1组(75mGy)照射后,细胞凋亡指数(AI)呈增加趋势,但与假照组(0mGy)比差异不显著(P>0.05)。D2组(4Gy)、D1+D2组细胞凋亡指数均明显增加,与假照组(0mGy)比差异显著(P<0.010。而D1+D2组与D2组比较,D1+D2组细胞凋亡指数增加更为明显,有统计学差异(P<0.05)。结论75mGy照射对小细胞肺癌可能有一定的促细胞凋亡作用;大剂量照射及低剂量联合大剂量照射对小细胞肺癌均有促细胞凋亡作用,低剂量辐射与大剂量辐射有协同促小细胞肺癌细胞凋亡的作用。 Objective To explore the effect of low dose radiation (LDR) on apoptosis of transplantation tumor tissues of human small cell carcinoma (NCI-H466)-bearing nude mice. Methods Tumor (NCI-H446)-bearing nude mice transplantation tumor tissues were radiated by whole body X-ray. The apoptosis of transplantation tumor tissues of human small cell carcinoma (NCI-H466) was determined by TUNEL. Results The apoptotic index in 75 mGy group were slight increased, but without significant difference comparing to that in sham-radiated group (0 mGy) (P>0.05). The apoptotic index in 4 Gy group and that in 75 mGy & 4 Gy group were all significantly increased comparing to that in sham-radiated group (P<0.01). But the apoptotic index in 75 mGy & 4 Gy group was significantly increased comparing to that in 4 Gy group (P<0.05). Conclusions Low dose radiation might promote apoptosis of transplantation tumor cells to some extent. High dose radiation and low dose united high dose radiation could all promote apoptotic function of small cell carcinoma.
