金属硫蛋白-1与p53基因在燃煤型砷中毒患者口腔黏膜脱落细胞中的表达 被引量:8

摘要 目的观察金属硫蛋白-1(MT-1)、p53基因、雌激素受体(ERa)基因和甲胎蛋白(AFP)基因在燃煤型砷中毒患者口腔黏膜脱落细胞中的表达。方法收集贵州省兴仁县燃煤型砷中毒患者及非病区正常人口腔黏膜脱落细胞,提取mRNA,以荧光实时定量PCR(RT-PCR)方法检测与肝硬化及皮肤癌相关的MT-1及p53基因等表达。结果燃煤型砷中毒患者口腔黏膜脱落细胞MT-1基因表达与对照组比较明显减少(0.83±0.15、3.86±0.37),抑癌基因p53明显增加(4.35±2.79、0.90±0.58),ERa基因和AFP基因无变化。结论砷中毒患者口腔黏膜MT-1基因表达减少,抑癌基因p53表达增加,可作为燃煤型砷中毒患者皮肤及肝脏病变监测的敏感指标之一,RT-PCR技术可广泛用于砷中毒的防治、监测和科研。 Objective To observe the hepatic disease and skin cancer related genes,metallothionein-1(MT-1), p53, estrogen receptor-α(ER-α), α-fetal-protein(AFP), expression in cells exfoliated from buccal mucous membrane of coal-burning arsenism patients. Methods These cells were collected by washing with 0.9% NaCl and mRNA were extracted from both arsenic patients and normal controls. Real-time PCR(RT-PCR) was employed to detect the change of related genes expression. Results MT-1 was found significantly lower and p53 was found overexpressed in arsenic patients than that in control, while other genes expressions maintained same as those of controls. Conclusions Expression of MT-1 and p53 gene in buccal mucous membrane cells may be regarded as sensitive molecular markers for skin and hepatic pathologic changes in arsenic patients. RT-PCR can be used as an efficient tool in arsenic diagnosis and prevention.
