肾3号方对IgA肾病大鼠的作用机制探讨 被引量:1

摘要 目的:观察肾3号方对IgA肾病(IgAN)大鼠的作用机制。方法:以牛血清球蛋白(bovinegammaglobu lin,BGG)免疫Lewis大鼠建立IgA肾病大鼠模型。设正常对照组、IgAN组、IgAN+肾3号方治疗组。IgAN+肾3号 方治疗组于第4周末~第8周末每日灌服肾3号方水提取液。蛋白质印迹法(Westernblot)、免疫组化检测转化生长 因子(TGF-β1)表达,免疫组化测IgA肾内沉积,进行尿红细胞、肾组织学检测。结果:与IgAN组比较,IgAN+肾3号 方治疗组8周末无明显血尿(P<0.01),肾小球系膜增生程度减轻,免疫组化IgA在肾小球沉积明显减轻(P<0.01), 免疫组化TGF-β1肾小球阳性显著减轻(P<0.05),WesternblotTGF-β1明显减少(P<0.05)。结论:肾3号方能减 轻IgAN鼠血尿,减轻肾组织病理损害,减轻肾小球IgA沉积和肾TGF-β1的表达和产生。 Objective:To observe the mechanism of Shen- No.3-Fang (S3F) on rat IgA nephropathy.Methods:Rat IgA nephropathy (IgAN) was induced by continuous oral immunization with bovine gamma globulin (BGG) in the drinking water for eight weeks, and followed by intravenous injection of BGG 1 mg daily for three days.14 rats were randomly divided into three groups: 4 for control, 5 for IgAN,and 5 for IgAN+S3F.S3F decoction was fed to rats every day from the end of the 4th week to the end of 8th. RBC in urine and pathological changes were examined by light microscope, IgA deposited in kidney tissue was tested by immunofluorescence and expression of transforming growth factor beta 1(TGF-β 1) in kidney was examined by Immunohistochemistry and Western blot.Results:Compared with control group,IgAN rats developed markedly hematuria, more profound mesangial proliferation and mesangial expansion, more severe and diffuse IgA deposition in glomeruli,more intensive expression of TGF-β 1 in glomeruli and interstitium,and significantly increased kidney TGF-β 1 by Western blot. Compared with untreated IgAN rats,there were significant decrease in hematuria, reduction of IgA deposition,and weaker expression and production of TGF-β 1 in kidney in IgAN+S3F treated rats.Conclusion:S3F can reduce the hematuria , the expression of TGF-β 1 in kidney and the deposition of IgA in glomerulus ,also can relieve the lesion of renal histopathology of IgAN rats.
