链脲佐菌素-糖尿病大鼠视网膜微血管病理改变 被引量:6

An investigation of the pathology of the retinal microvascular network in rats with diabetes induced by streptozotocin

摘要 目的 :观察STZ 糖尿病大鼠视网膜微血管的早期病理变化。方法 :4 5只SD大鼠被随机分为糖尿病组 (DM)和对照组 (CON) ,用链脲佐菌素 (streptozotocin ,STZ)诱导SD大鼠建立糖尿病动物模型 ,分别饲养 3个月和 6个月。各组大鼠到期后 ,取眼球 ,采用视网膜消化铺片PAS染色、细胞图像分析及透射电镜技术进行早期糖尿病视网膜微血管形态学检查 ,记数微血管细胞数和无细胞血管数目 ,观察微血管及细胞超微结构。结果 :糖尿病视网膜早期微血管周细胞数目逐渐减少 ,无细胞血管增多。超微结构显示微血管细胞结构异常 ,基底膜变性、增厚。结论 :糖尿病视网膜病变早期存在微血管细胞病变、基底膜变性增厚及无细胞血管形成的改变。临床应重视糖尿病视网膜病变 (DR)的早期防治 ,改善微血管循环。 Objective:One of the earliest changes observed in retinal microvessels in diabetic retinopathy is the selective loss of intramural pericytes. The purpose of this study was to observe the dynamic changes in the pathology of retinal microvascular morphology in the early stage of diabetic retinopathy.Methods:Forty five SD rats were randomly divided into a control group (CON) and diabetic mellitus (DM) group. Diabetes was induced in animal models by the injection of streptozocin (60 mg/kg) through the abdomens of 30 SD rats. Blood sugar levels were tested after 48 hours. Animals whose blood sugar exceeded 16.7 mmol/L were regarded as diabetic models. The rats were sacrificed at 3 months and 6 months of age and the eyeballs were fixed by 4% polyformaldehyde within 24 hours. The retina was then isolated from the optic disc and retinal trypsin digests were prepared. After PAS staining,the number of pericyte and epithlium cells and acelluar capillaries were counted in the cell image analysis system (CIAS). Thickening of the basement membrane of the blood vessels and cell images were observed with electron microscopy.Results:The respective numbers of pericyte and epithelium cells in the visual field in the control group were (6.71±0.45) and (22.47±3.16) at 3 months and (6.52±0.40) and (22.56±3.11) at 6 months. For the DM 3 group,the respective numbers were (6.17±0.45) and (23.10±3.00). For the DM 6 group,the respective numbers were (5.66±0.60) and (23.45±2.99). Compared to the CON group,the number of pericytes decreased gradually in the DM 3 group (P<0.01) and more obviously in the DM 6 group. There were no obvious changes in the number of epithlium cells in each group at the different periods. The number of acelluar vessels in the visual field were (3.6±3.2) and (5.1±3.3) at 3 months and 6 months,respectively. The number for the DM 3 group was (16.4±3.9) and the number was (29.7±5.9) for the DM 6 group. Compared to the CON group,the number of acelluar vessels increased gradually in the DM group (P<0.01). The ultrastracture of the retinal microvascular cells was abnormal and the retinal capillary basement membrane thickened and degenerated.Conclusion:In the early stage of diabetic retinopathy,there were changes in the pathology of retinal vessel cells,the basement membrane thickened and degenerated and acelluar capillaries formed.
