摘要
目的:制备家兔肝抑素纯品,检测其生物活性、纯度和相对分子质量。方法:应用超速离心法和酒精沉析法从家兔肝中提取肝抑素制成粗品,采用SephadexG-50凝胶过滤层析法和DEAE-cellulose阴离子交换层析法制备肝抑素纯品,采用SDS聚丙烯酰胺电泳和扫描定量分析来检测肝抑素的纯度和相对分子质量,用3H-TdR掺入液闪测定(CPM)法体外检测肝抑素样品的生物活性。结果:提取的肝抑素纯品的纯度达88.13%,相对分子质量为13500,该纯品对家兔再生肝细胞在体外的增殖有明显的抑制作用。结论:提取的肝抑素为纯品,它在肝再生过程中起重要调控作用。
Objective:To purify rabbit hepatic chalone(HC),and investigate its biological activity,purity and molecular weight.Methods:HC crude extract was prepared from rabbit livers by ethanol precipitation a nd ultracentrifugation.The Sephad ex G -50gel chromatography and DEAE -cellulose anion exchange chro matography were perforened to purify HC sample.The SDS -PAGE was used to a ssay the purity and molecular weight of HC.The 3 H -TdR incorporation(Fluid scintillation,cpm)was applied to the biological activity of HC sample in vitro.Results:The purified HC is 88.13%of purity an d 13500of molecular weight.This pur ified HC showed remarkable inhibito ry effect on proliferation of regenerating hepatocytes in vitro.Conclusion:The HC in this study was confirmed to b e purified products which exerted an important regulating effect on li ver regeneration.
