摘要
以安眠酮为内标,甲醇-水(72:28,v/v)为流动相,在Shim-PackCLC-ODS(150mm×6mm,5μm)柱上,研究并建立了测定血瘀证大鼠微量血清中川芎嗪浓度的高效液相色谱法。200μl血清以三氯甲烷萃取,浓缩后以甲醇定容,进样分析。川芎嗪的检测限为2.2ng(S/N=3);最低检出血清浓度为0.068μg/ml。线性范围0.22~6.71μg/ml,r=0.9994。方法平均回收率为98.29±2.98%;日内及日精密度RSD值均<5.75%。本方法具有专一性强、灵敏、简便而快速的特点。内源性物质及制动物模型所用药物均不干扰测定。用本方法测定了正常组及血瘀组大鼠的药代动力学参数。
A RP-HPLC method was established for the determination of tetramethylpyrazine(TMP) in serum of blood stasis rat. Methaqualone is used as the internal standard. TMP is extracted from serum with chloroform and evaporated to dryness. The mobile phase consists of a mixture of CH_3OH-H_2O (72:28. V/V). The analytical columm is Shim-pack CLC-ODS (150mm ×6mm. 5μm). The TMP is detected at 280nm. The recovery is 98. 29±2. 98%. The RSD of within-day and day-to-day are less than 5. 75%. The method is applied to pharmacokinetic study of health and blood stasis rats.
