4—NP摩尔吸光系数的影响因素及对ALP测定结果影响的探讨

摘要 本文ALP测定中监测物4—NP摩尔吸光系数(e)影响因素进行了探讨。六台生化仪测定同—ALP试剂盒反应液中4—NPe′,结果分别为:15.98、16.72、17.00、16.72、17.62。同一仪器测定四种不同 ALP试剂盒中 4—NPe′,结果分别为:16.90、17.38、17.72、16.11。六台仪器,同一试剂盒测定三种浓度质控血清ALP,按试剂盒中e计算,各仪器结果间有显著差异(P>0.01),按各仪器实测e′计算,各仪器间结果差异不显著(P>0.05)。同一仪器,四种不同试剂盒,测定12个血清标本ALP,按各试剂盒中e计算,各试剂盒间测定结果有显著差异(P<.01),按各试剂盒实例e′计算,各试剂盒问结果差异不显著(P>0.05)。建议在酶测定试剂盒中配上被监测的的标准液,可克服因不同仪器不同试剂使用同一e而导致的酶测定的系统误差。 We have studied the factors that influence the molar absoptivity of 4-Np.ε'Of4-NP determined by six different inst ruments with the same kit for ALP is 15.98,16.72, 16.06, 17. 00, 16.27, 17. 62, respectively, and by the same instrumentwith four different kits for ALP is 16.90, 17. 38, 17. 72, 16. 11, respectively.Seruma of three different ALP concentration wre measured by six instruments withthe same kit for ALP. Differences of results according to εof kits by the sixinstruments are well important (P<0. 01), and according to ε'which is determined bydifferent instruments are not (P>0. 05). Furthermore we have examined ALP in ?2clinical sampales by the same intrument with four different kits for ALP. If theresults depend on εof kits, the differences among the different kits are well obvious(P<0. 01), however if the results depend upon the caculation of actual ε'withdifferent kits, the differences are not well significant (P>0.05). In a conequence,we sugget that standard material be contained in kit for enzymatic detection in orderto get rid of sytematic error caused by same ε introduced in kit.
