摘要
目的 建立田参氨基酸胶囊中1 0种氨基酸的含量测定方法。方法 以2 ,4 -二硝基氟苯(FDNB)为衍生化试剂,将胶囊中的氨基酸定量衍生化,用RP -HPLC测定其含量。衍生化条件为在碱性溶液中6 0℃反应1h。色谱条件为ODS柱,甲醇-醋酸盐缓冲液梯度洗脱,36 0nm处检测。结果 5 0min内氨基酸均可较好地分离。衍生化产物性质稳定;1 0种氨基酸在各自浓度范围内线性关系良好(r≥0 . 9998) ,平均空白加样回收率均为1 0 0 .1 %~1 0 1 . 1 %,RSD =0 . 70 %~1 . 0 8%;日内及日间精密度均小于1 .9%。结论 方法准确,简便,费用低,可用于田参氨基酸胶囊的质量控制。
OBJECTIVE To develop a method for determination of amino acids in Tianshen Anjisuan capsule. METHODS Derivated by 2,4-dinitrofluorobenzene, 10 kinds of DNP-amino acids in the capsule were analyzed by RP-HPLC with an ODS column as stationary phase, methanol/acetate buffer as mobile phase,360 nm as detective wavelength.RESULTS All amino acids were separated (resolution≥1.3)within 50 min, with average recoveries of 100.1%-101.1% and RSD of 0.70 %-1.08%.The RSD of within-day and between-day were less than 1.9%. CONCLUSION This method is accurate, simple, cheap, and can be used for determination of amino acids in Tianshen Anjisuan capsules.
