摘要
目的:研究参附注射液(SF)对大鼠心肌的保护效应与HSP70 的关系。方法:采用整体缺血再灌注(I/R)模型,35只大鼠随机分为假手术组(C组)、I/R组2组、SF组2组,心肌缺血4 0min再灌注30min或12 0min检测血流动力学参数,心肌MDA含量、SOD活性、HSP70的表达及超微结构。结果:与I/R组比较:SF组的LVSP、±dp/d/max均升高而LVEDP降纸(P<0 .0 5 ) ;I/R12 0min后SF组SOD活性增加、MDA含量减少(P <0 .0 5 )、HSP70 的表达增强(P <0 .0 5 ) ;电镜显示SF组的心肌损伤程度均轻于I/R组。结论:参附对I/R心肌的保护效应与其抗氧自由基及抗脂质过氧化反应作用和促进HSP70
Objective:To study the relationship between the protective effect of shenfu parenteral solution (SF) on myocardium in rats and HSP 70 .Methods:To establish general ischemia reperfusion(I/R) model.Thirty-five healthy white rats were divided into five groups randomly:sham operation group(group C),I/R30 group (including 2 groups),SF group(including 2 groups).After myocardial ischemia lasting for 40min,then reperfusing for 30min or 120min.To detect hemodynamic parameter,myocardium MDA content,SOD activity,HSP 70 expression and ultrastructure were examined.Results:Compare with I/R group:LVSP and ±dp/d/max rose up,however,LVEDP decreased( P <0.05) in SF group.After mycocardial ischemia lasting for 120min,SOD activity in SF group increased,MDA content decreased( P <0.05) and HSP 70 expression reinforced( P <0.05).Electron microscope showed myocardium injury was lighter in SF group than in I/R group.Conclusion:The SF protective effect on myocardium was connected with its anti-oxygen-derived free radicals and anti-lipid peroxiaation response,as well as with acceleration of HSP 70 expression.
