摘要
目的:建立脾胃虚寒型胃溃疡动物模型,为临床研究提供实验方法。方法:采用饮食失节与改良Okabe法制备脾胃虚寒型胃溃疡动物模型,观察溃疡发生率、耐寒性(游泳时间)、溃疡面积及血清胃泌素(GAS)含量的变化。结果:全部制模动物都出现胃溃疡,溃疡发生率10 0 %。模型组动物的游泳时间较空白对照组动物明显缩短(P<0 .0 1) ,附子理中丸组接近空白对照组。模型组(证+型)溃疡面积为(1.4 36±0 .5 0 5 ) m m2 ,而空白对照组和模型组(证)均为0 (P均<0 .0 1)。血清GAS含量模型组较空白对照组明显减少(P<0 .0 1) ,附子理中丸组接近空白对照组。结论:脾胃虚寒型胃溃疡动物模型与临床脾胃虚寒证和胃溃疡的病理过程类似。可重复性高。
Objective: To establish a rat model with gastric ulcer of deficiency cold of spleen and stomach type(脾胃虚寒型) to offer an experimental method for clinical study. Methods: The rat model was made by dietary ignorance and modified Okabe method. The incidence of gastric ulcer, cold endurance (swimming time), area of ulcer, changes of serum gastrin (GAS) content were observed. Results: Gastric ulcer occurred in all the rat models (100%). The swimming time in the model group was much shorter than that of the blank control group (P<0.01), and the time in the Fuzi Lizhong pill(附子理中丸) group was close to that in the blank control group. GAS content of the model group was much lower than that of the Fuzi Lizhong pill group(P<0.01). The ulcer area of the model group(syndrome+type) was (1.436±0.505)mm 2 and those of the blank control group and model group(syndrome) were 0 (both P<0.01). The serum GAS content in the model group was significantly lower than that in the blank control group (P<0.01), and the content in the Fuzi Lizhong pill group was close to that in the blank control group. Conclusion: The pathological process of rat model with gastric ulcer of deficiency cold of spleen and stomach type is similar to that of its clinical counterpart. The repeat rate of the model is high.
