地龙2号对大鼠肝纤维化α-SMA、TGF-β1、MMP-13及TIMP-1蛋白表达的影响 被引量:18

摘要 目的研究地龙2号对肝纤维化大鼠肝组织α_SMA、TGF_β1、MMP_13及TIMP_1蛋白表达的影响。方法采用四氯化碳皮下注射诱导大鼠肝纤维化模型,5 2只雄性Wistar大鼠随机分成6组正常组、阴性对照组、模型组、阳性对照组、地龙2号大剂量组和地龙2号小剂量组,8周末用免疫组织化学染色法检测肝组织中α_SMA、TGF_β1、MMP_13及TIMP_1蛋白表达情况。结果与模型组相比,地龙2号组肝纤维化程度明显减轻,α_SMA、TGF_β1、及TIMP_1蛋白表达均显著降低,MMP_13表达显著升高。结论地龙2号抑制大鼠肝脏纤维组织的形成可能与其降低α_SMA、TGF_β1、TIMP_1蛋白表达并促进MMP_13蛋白表达有关。 Objective To investigate the effects of No.2 of Earthworm's extractions (EE2) on the degree of hepatic fibrosis and the proteinic expressions of α_SMA,TGF_β1,MMP_13 and TIMP_1 of hepatic fibrosis in rats.Methods Fifty_two male Wistar rats were randomly divided into 6 groups: normal group (N group, 6 rats), normal control group (NC group, 6 rats), model group (M group, 10 rats), positive control group (PC group, 10 rats), high dose of EE2 group (Eh group, 10 rats) and low dose of EE2 group (E1 group, 10 rats). The rats of M, PC, Eh and E1 group were with mixture of CCl_4 and peanut to induce liver fibrosis, sc twice a week. The Eh or E1 group was also treated with high dose or low dose of EE2, respectively 0.05 g/kg or 0.025 g/kg, ig once a day; the PC group was treated with colchicines, ig once a day; the M and NC groups were given singly_distilled water, ig once a day; and the NC group was only administrated with peanut oil, sc twice a week. At the end of 8 weeks all the survived rats were sacrificed. The pathological changes of liver were observed by H_E staining. The proteinic expressions of α_SMA,TGF_β1, MMP_13 and TIMP_1 were observed by the method of immunohistochemistry. Results Compared with controls,The proteinic expressions of α_SMA、TGF_β1、and TIMP_1 were significantly lower in the Eh and E1 groups that those in the M group, while that of MMP_13 was higher in the Eh and E1 group.Conclusion EE2 could inhibit obviously the formation of liver fibrosis in the rats induced by CCl_4, which might relate to decreasing the expression of α_SMA、TGF_β1 and TIMP_1 and to increasing that of MMP_13.
