骨碎补提取液对兔骨髓基质细胞增殖的影响 被引量:25

摘要 为研究中药骨碎补提取液对兔骨髓基质细胞增殖作用的影响,探讨接骨续筋中药促进骨折愈合的机制,取新西兰大白兔骨髓液,经FICOIL淋巴细胞分离液分离后行原代培养,以P1代细胞为研究对象,分为5组分别进行培养,MTT法测定其生长曲线,测量其分裂指数,行HE染色观察细胞形态,对测量的结果行统计学分析。结果显示条件培养组与完全培养基组无显著性差异,不同浓度骨碎补组均与完全培养基组有显著性差异。表明高浓度骨碎补提取液对于兔骨髓基质细胞的增殖有抑制作用,较低浓度的骨碎补提取液对兔骨髓基质细胞的增殖有促进作用,条件培养基对兔骨髓基质细胞的增殖无明显作用。 In order to study the effect of 'GUSUIBU' (Drynaria baronii) extracted liquid (GEL) on rabbit marrow-stromal-cell proliferation (RMSCP) and to explore the fracture-healing-stimulating mechanism of the fracture-rejoining and soft-tissue-restoring TCD, the bone marrow of New Zealand white rabbit was separated by FICOIL lymphocyte separation liquid and primarily cultured and the P1 - generation cells were used as the subjects, divided into five groups and cultured, respectively; the growing curves and the division index were measured by MTT method; the cell morphology was observed through HE staining; the results of measurement were statistically analyzed. The results showed that the combinatively-induced group had no significant difference from the complete medium group, while the variedly-concentrated GEL groups had significant differences from it, suggesting: a. high-concentrated GEL could inhibit RMSCP; b. lower-concentrated GEL could stimulate RMSCP; c. the conditioned medium had no obvious effect on RMSCP.
