胰腺癌组织中高迁移率族蛋白1和基质金属蛋白酶2的表达研究 被引量:8

摘要 目的:研究胰腺癌组织中高迁移率族蛋白1 ( high mobility group box 1,HMGB1)及基质金属蛋白酶2 ( matrixmetalloproteinase 2, MMP 2)蛋白表达并探讨其与癌分化程度、肿瘤大小、浸润、淋巴及血道转移的关系。方法:用免疫组织化学SP 方法,检测73 例胰腺癌组织、30例癌旁组织HMGB1 及MMP -2 蛋白表达。结果:HMGB1和MMP 2在胰腺癌中的阳性表达率分别为76. 7% ( 56/73 )和64 4% ( 47/73 ),在癌旁组织中分别为20 .0%(6/30)和23 3%(7/30), 两者在癌组织与癌旁组织间表达差异均有统计学意义,P<0. 01。HMGB1 和MMP- 2 的阳性率与癌分化程度无关,P>0 .05;与肿瘤的大小、浸润、淋巴及血道转移呈正相关,P<0 .05或<0 .01;HMGB1 和MMP -2 在胰腺癌组织中表达呈正相关,P< 0. 01。结论:HMGB1 和MMP- 2 在胰腺癌生长、侵袭和转移过程中起着重要的作用, OBJECTIVE: To investigate the expressions of high mobility group box 1( HMGB1) and matrix metalloproteinase 2 (MMP-2) in human pancreatic carcinoma and to explore their relationship to tumor size, differention, regional invasion, lymphatic and blood stream metastasis.METHODS:The immunohistochemical S-P method was used to detect the expressions of HMGB1 and MMP-2 in 73 pancreatic cancer tissues and 30 juxtacancerous tissues.RESULTS:The positive rates of HMGB1 and MMP-2 in pancreatic cancer were 76.7%(56/73) and 64.4%(47/73),respectively, 20.0%(6/30) and 23.3%(7/30) in juxtacancerous tissues, respectively, and both had significant expression difference between pancreatic carcinoma tissues and juxtacancerous tissues,P<0.01. The expressions of HMGB1 and MMP-2 were markedly correlated to tumor size, regional invasion, lymphatic and blood stream metastasis, P<0.01, but not to the differentiation degree, P>0.05, in pancreatic cancer. The HMGB1 expression was positively correlated to MMP-2 expression in pancreatic carcinoma.CONCLUSIONS: The strong expressions of HMGB1 and MMP-2 in pancreatic cancer may play a great role in the progression of pancreatic tumor growth, invasion and metastasis. Examination of expressions of the two proteins might be helpful to judging biological behavior and prognosis of pancreatic carcinoma.
