摘要
获得高质量的植物总RNA比较困难,RNA的提取方法会因植物材料的不同而又有较大差异.棉花幼根含有大量多酚、多糖、单宁和其它多种次生代谢物,因此RNA提取难度较大.本文报道了棉花根样的高效获取方法和经研究改进的棉根RNA热酚提取法,该法可获得高质量棉根总RNA,并能成功进行反转录,制备cDNA.
It is relatively difficult to extract quality total RNA whose extracting methods vary depending on different plant materials. Seedling roots of cotton contain polyphenol, polysaccharide, tannin and other secondary metabolites in great quantity thus making RNA extraction difficult. The paper reports the high-performance sampling method for sampling cotton roots and the modified hot phenol method for extracting total RNA from seedling roots of cotton evolved from researches, which is capable of obtaining quality total RNA from cotton roots, and conducting its reverse transcription to prepare cDNA.
