摘要
目的:研究开发适用于小鼠胚胎干细胞(EmbryonicStemCell,ES细胞)的低温贮存试剂。方法:以DM SO为主要低温贮存保护剂检测不同多羟基化合物对小鼠胚胎干细胞低温贮存后存活率和多向分化能力的影响。结果:在含有DMEM(高糖)、10 %DMSO、10 %FBS、10mg/mL抗坏血酸、0 .18mg/mL肌醇、0 .4 4mg/mL叶酸的低温保护剂中补充3 4 .2 3mg/mL海藻糖能够有效维持小鼠胚胎干细胞的自我更新能力和多向分化能力。
Objective:To develop the cryoprotective additives used for cryopreservation of mouse embryonic stem(ES) cell. Methods: The forming rates of mouse ES colonies and embryod bodies(EBs) in cryopreservation experiment were checked up by adding three polyols respectively into cryoprotector including DMEM(high glucose), 10%DMSO, 10%fetal bovine serum, 10mg/mLVc, 0.18mg/mL Inositol,0.44mg/mL Folic acid. Results:the cryoprotector added with 34 23mg/mL trehalose could effectively preserve self-renewing ability and pluri-potency of mouse ES cells.Conclusion: A new cryoprotectivery reagent with DMSO and trehalose and so on for mouse's embryonic stem cell cryopreservation has been developed.
