慢性氟中毒大鼠肾脏损害机理及部位研究 被引量:8

摘要 用30、60和120mg/L含氟水饲养大鼠12周。结果发现:尿r—GT活性和尿蛋白含量显著升高;肾MDA含量显著升高,肾GSH含量和肾GSH-Px活性显著降低;肾损害指标与肾脂质过氧化指标存在显著相关关系。肾线粒体和微粒体MDA含量显著升高,其各自标志酶SDHase和G-6-Pase活性显著降低,而且各亚细胞器MDA含量升高分别与各自标志酶活性降低存在显著负相关关系.结果提示:慢性氟中毒引起肾脏近曲小管和肾小球损害;损害机理之一是脂质过氧化,慢性氟中毒可致肾细胞线粒体和微粒体发生脂质过氧化损害作用. The rats drank water supplemented with 30,60,120 mg/L fluorine for 12 weeks.the following results were found:The urinary r-GT activities and urinary protein contents were markedly increased.The significances were found in the increases of renal MDA contents and in the decreases of renal GSH contents and renal GSH-Px activities.The changes of renal damage induces were significant correlations with the changes of renal lipid peroxidation indices.The contents of MDA in renal mitochondria and microsomes were markedly incteased.The activities of SDHase in renal mitochondria and G-6-Pase in renal microsornes were significantly reduced.The increases of MDA contents in two subcellar frations were significant ntgative correlations with the decreases of their enzymes activities respectively.The results suggest that the proximal tubules and glomerali of kidney were damaged in rats poisoned chronically by fluorine,the one of mechanisms of renal damages was lipid peroxidation the damage effects of lipid peroxidation in renal mitochondria and microsomes of rats were induced by chronic fluorosis.
