摘要
目的 探讨免疫组织化学反应不同预处理方法对反应结果的影响。方法 对5 0例经10 %中性福尔马林液固定过的组织,经脱水、石蜡包埋,制成切片,分别采用3种不同方法作预处理后,进行免疫组织化学(S P法)染色。结果 石蜡组织切片免疫组化反应预处理,采用中温水浴抗原修复法,其阳性表达率为5 2 .6% ,脱片率为2 .0 % ;采用高压锅抗原修复法,其阳性表达率为47.4% ,脱片率为2 0 .0 % ;采用水沸抗原修复法,其阳性表达率为3 9.5 % ,脱片率为12 .0 %。结论 免疫组化反应采用中温水浴抗原修复法作预处理,石蜡组织切片阳性表达率较高压锅抗原修复法提高了5 .2 % ,P <0 .0 5 ,较水沸抗原修复法提高了13 .1% ,P <0 .0 5 ;采用中温水浴抗原修复法石蜡组织切片脱片率较高压锅抗原修复法降低了18.0 % ,P <0 .0 1,较水沸抗原修复法降低了10 .0 % ,P <0 .0 1。预处理采用中温水浴抗原修复法,即提高了免疫组织化学反应阳性表达率,又提高了制片质量,操作方法易于掌握,是1种理想的免疫组化反应预处理方法,值得推广。
Objective To explore the influence to the reaction result that caused by the different pretreatment method of immunity organization chemical reaction.Methods Select 50 organization samples that fixed by 10% neutral formalin,dehydrate them,pack them with paraffinwax and chip them.Then use three different methods to pretreat them,finally use S-P method to do the immunity organization chemical dyeing.Results Prerteat organization chip,using the middle temperature water bath antigen repairing method,the positive expressing rate is 52.6% and the unfalling rate is 2.0%;using the pressure cooker antigen repairing method,the positive expressing rate is 47.4% and then unfalling rate is 20.0%;using the water boiling antigen repairing method,the positive xpressing rate is 39.5% and the unfalling rate is 12.0%.Conclosion The positive expressing rate of the middle temperature water bath antigen repairing method is 5.2% higher than that of the pressure cooker antigen repairing method(P<0.05) and is 13.1% higher than that of the water boiling antigen repairing method(P<0.05).The organization chip unfalling rate of the middle temperature water bath antigen repairing method is 18.0% lower than that of the pressure cooker antigen repairing method(P< 0.01) and is 10.0% lower than that of the water boiling antigen repairing method(P<0.01).The middle temperature water bath antigen repairing method,increase the positive expressing rate of the immunity organization chemical reaction and improve the chip quatity.It's easy to maste and is widely used.It's a good pretreatment method of immunity organization chemical reaction and is worth being expanded.
