摘要 目的:运用正交实验优化去甲斑蝥素微囊的制备工艺。方法:采用凝聚法制备了去甲斑蝥素微囊,以包封率、微囊强度为指标,设立总的评估指数,选取海藻酸钠浓度、壳聚糖浓度、氯化钙浓度和去甲斑蝥素用量4 个因素,每个因素选取3 个水平,选用L9(34)表进行正交实验优化制备工艺。结果:4个因素中,壳聚糖浓度对评估指数的影响最大(P<0.01),其次是氯化钙和海藻酸钠浓度(P<0.05),去甲斑蝥素对评估指数的影响没有统计学意义(P>0.05)。按照优化方案进行正交实验复核实验表明,药物包封率、微囊机械强度分别为96.35%、98.00%。结论:本法工艺简便、稳定,具有应用前景。 OBJECTIVE To study the new preparation of norcantharidin(Nctrd )chitosan-alginate microencapsules.METHODS Norcantharidin chitosan-alginate microencapsules(Nctrd-CAM)were prepared by coacervation technology.Four factors,including alginate,chitosan,CaCl_2 and Nctrd,and three levels for each factor that may affect the formulation of microencapsules were selected and arranged in an L_9(34) orthogonal experimental table. The evaluating indicator(EI%),calculated according to drug encapsulated efficiency(EE%) and strength of microencapsules(MS%),was introduced as a total index for the microencapsules formulation.RESULTS The four factors affected EI differently.Chitosan behaved as the maximum(P< 0.01),followed alginate and CaCl_2(P< 0.05).However,the Nctrd did not influence EI(P> 0.05).The orthogonal experiment was checked up and the result indicated that EE% and MS% were 96.35% and 98.00% respectively.CONCLUSION The preparation procedure is simple and reproducible.
