摘要 通过研究FK506在体外对人淋巴细胞趋化反应的影响。发现在FK506的浓度为0.5ng/ml时,对蛋-亮-脯氨酸甲酯多肽(FMLp-ME)、酵母多糖激活血清(ZAS)和趋化性胆汁(chemotacticbile,CB)引起的淋巴细胞趋化反应的抑制率分别为47.3%、32.2%和48.9%;对经PhorbolMyristateAcetate(TPA)激活的淋巴细胞,FK506的抑制作用更强,抑制率分别达71.2%、55.4%和69.6%。FK506也抑制TPA激活的淋巴细胞产生趋化因子。比较经不同处理的淋巴细胞培养上清液引起的白细胞趋化反应,发现TPA组与TPA+FK506组间存在显著性差异(淋巴细胞趋化反应P<0.001、单核吞噬细胞P<0.0005、中性粒细胞P<0.0005),而对未激活的淋巴细胞产生趋化因子无影响。可见对淋巴细胞趋化反应的抑制可能是FK506作用的一种方式。 AbstractAdams D. Theinhibitory activities of FK506 in concentration of 0.5ng/ml on the lymphocyte chemtaxis stimu-lated by FMLp-ME,ZAS or CB were 71.2%,55.4%and.69.5%respecticvely,It showed adose dependent pattern of inhibitory activity of FK506,i.e. the higher concentration of FK506,the stronger inhibition of chemotaxis of lymphocytes.In comparison with above data,markedhigh inhibitory activities were observed in the lymphocytes prestimulated by TPA with the in-hibitory rate of 71.2%,55,4%and 69.6% separately,A significant difference of chemotaxis forvarious cell type was observed in the supernatants of lymphocyte culture activated by TPA aloneor TPA with FK506(lymphocyte P<0.0O1;moncyte-macrophage P<0.0005; Neutrophil P<0.0005 ).No difference of chemotaxis in supernatants of unstimulated lymphocytes was re-vealed between FK506 and control.It is suggested that the therapeutic mechanism of FK506might be by means of inhibition of the chemotaxis of lymphocyte.
