摘要
目的 探讨血小板P选择素(P selectin)在哮喘患者中的表达及其临床意义。方法 体外分离13名健康对照者,37名哮喘急性发作患者,2 2名哮喘缓解者的血小板和血浆,用流式细胞仪对血小板进行P selectin测定,用放射免疫分析法对血浆血栓素B2 (TXB2 )、6- 酮前列腺素F1α(6 - K -PGF1α)进行测定。同步测定所有参试者肺功能,分析与P selectin表达的关系。结果 哮喘急性发作者P selectin、TXB2 、6 - K PGF1α均较健康对照者明显升高(P <0 .0 5 ) ,当经过治疗处于缓解期时,P selectin、6- K- PGF1α较健康对照组无明显差别(P >0 .0 5 )。P -selectin的表达水平与FEV1占预计值百分比(FEV1%Pred)成显著负相关(r =- 0 .5 83,P <0 .0 1)。结论 哮喘急性发作患者有P- selectin的高表达,表达水平与肺功能成负相关,P- selectin在哮喘的发病中起着一定的作用。
Objective To investigate the expression of P-selectin and its clinical significance in asthma. Methods Platelets and plasma were isolated from 13 normal subjects,37 patients with acute asthma and 22 stable patients,the expression rate of P-selectin was measured by flow cytometry,the plasma thromboxane B_2(TXB_2) and 6-keto-PGF_ 1α(6-K-PGF_ 1α) were determined by radioimmunoassay. Meanwhile the pulmonary function of all participants was evaluated,the relationship between pulmonary function and P-selectin was analysed. Results The level of P-selectinTXB_2 and 6-K-PGF_ 1α were all significantly increased in patients with the acute asthma group as compared with the control group (P<0.05),but there was no difference between the stable patient group and the control group (P> 0.05). The expression of P-selectin in patients with acute asthma was negatively correlated with their FEV_1% Pred (r=-0.568,P<0.01). Conclusion These results suggest there is high level expression of P-selectin in patients with acute asthma,the level is negatively correlated with its pulmonary function, P-selectin has a potent effect on pathogenesis of asthma.
