光动力学疗法中低温对脑损伤的保护 被引量:2

摘要 目的该实验通过对正常Wistar大鼠脑组织进行光动力学治疗时,观察全身低温对正常脑组织是否有保护作用。方法将34只Wistar大鼠分成3组,低温组,正常组,对照组。所有3组均在显微镜下颅骨开窗,在激光照射前4h腹腔注射血卟啉衍生物,激光照射,对照组只注射血卟啉衍生物,不行激光照射。激光照射24h后灌注固定,以照射中心冠状切取脑组织,观察激光照射区形态学改变及测量皮层损害深度。结果低温组和正常温组均可见神经元损伤及片状出血,对照组未见损伤,低温组皮层损害的深度明显浅于正常温组(P<0.001)。结论提示轻度低温能减轻光动力学疗法时对正常脑组织的损害。 This study was designed to observe that the protection of whole body hypothermia in conjunction with photodynamic therapy (PDT) which was determined on normal rat brain. 34 wistar rats were divided into three groups. Hypothermic animals, normathermic group,control group. All of three groups, a 5-mm diameter craniectomy was performed under the microscope. Photofrin was injected intraperitoneally prior laser treatment. PDT was performed. Photofrin was only injected on the control group, and have no laser treatment. Cortica lesion depths were measured. and pathological evaluation was made at 24 hours post PDT. Lesions generated in hypothemic and normathermic groups showed similar pathology, characterized by coagulation necrosis and areas of hemorrhage, control group showed no pathology. But comparisons of the mean lesion depth of hypothermic and normathermic animals showed significant difference(P﹤0.001). the lesion depth of hypothermic animals were apparent shallow. [Conclusions] This results suggest that in vivo mild hypothermia may reduce damage to normal brain from PDT.
