卵龄及激活液中Ca^(2+)浓度对小鼠卵母细胞电激活效率的影响

摘要 采用宁波新科CRY-3细胞融合仪及其配备的镀银电融合槽(电极宽度为0.5mm)对影响电激活效率的两个因素——卵龄及激活液中Ca2+浓度进行了比较研究。结果表明:①卵母细胞的活化率随卵龄的增加明显升高,电刺激hCG后18~19h的卵母细胞可获得较高的卵母细胞激活率(71.67%),继续增加卵龄,卵母细胞激活率、卵裂率和囊胚发育率开始下降。②使用含Ca2+的激活液可获得较高的卵母细胞激活率,但Ca2+的浓度变化对激活效果无显著影响(P>0.05)。 In this study, different age and Ca2+ which may influence the effects of electric pulsing were compared. The results are as the following: ① Activation rates of oocytes were raised with the increase of their ages. The cleavage rate of oocytes at 18~19 h after hCG injection was clearly higher than others(71.67%). But the rates of activation, cleavage and development of blastocysts started to fall with the continue increase of ages. ② Ca2+ in electric medium was crucial to mouse oocytes activation when stimulated with electrical pulses. Oocytes were activated more easily in a medium with Ca2+ than in one without Ca2+ by electrical stimulations, but free calcium concentration in electric medium did not affect the activation efficiency of oocytes (P>0.05).
