摘要
对姬松茸菌丝进行了富集锌的研究。首先对姬松茸的耐锌和富锌特性进行了研究,结果表明,姬松茸的耐锌能力和富锌能力都很强,在锌浓度为5 0~2 0 0 0 μg/mL的固体培养基上,菌丝均能生长,当锌浓度超过30 0 μg/mL时,菌丝生长受到一定影响;采用液体深层培养,当锌浓度为2 0 0 μg/mL时,富锌率最高。实验中还对培养条件进行了优化,在培养温度2 5℃,pH 5 1,振荡速度15 0r/min ,2 5 0mL三角瓶装液5 0mL ,接种量15 %(v/v) ,培养基中锌添加量为2 0 0 μg/mL时,姬松茸菌丝的生物转化量及菌丝体富锌率最高,达到12 6 4 %。最后对锌的有机化进行了研究,结果表明,姬松茸对锌有生物转化能力,加入的锌并非机械地物理吸附在菌体表面,而是发生了有机的生物转化,形成了牢固结合的复杂的有机锌,其有机化程度在5 7%左右。
Accumulation of Zn in Agarics blazei Murill was determined and the Zn-resisting and Zn-accumulating characters of submerged culture Agarics blazei Murill were studied in this paper. The results showed that the capacity of both Zn-resisting and Zn-accumulating of Agarics blazei Murill was very high. The strain can grow in the solid medium with zine from 50 to 2000 μg/mL. However, the zine content above 300 μg/mL hampered its growth. When zine added to the liquid medium is up to 200 μg/mL, the hyphae have the most zine absorbance. It is also found that the Zn-recovery and the biomass of Agarics blazei Murill were the highest under the liquid culture conditions, that is, fermention temperature 25℃, stirring speed 150 r/min, inoculation size 15%, 250?mL shake flash containing 50?mL medium containing Zinc, pH5.1. In our study, the degree of organized-Zn was up to 57%.
