摘要
目的探讨瘦素、神经肽Y及其受体在慢性肾功能不全中的变化及意义。方法将40例W istar大鼠随机分为正常对照组(n=15)和慢性肾功能不全组(n=25),采用逆转录聚合酶链反应(RT-PCR)和免疫组织化学技术检测脂肪组织瘦素和脑组织瘦素受体(Ob-Ra、Ob-Rb)及神经肽Y(NPY)的基因及蛋白表达。结果慢性肾功能不全大鼠脂肪组织瘦素基因表达降低而蛋白表达增强,脑组织可见Ob-Ra和Ob-Rb的表达增强及NPY的表达降低。结论慢性肾功能不全大鼠存在明显的NPY、瘦素及其受体的代谢紊乱,过量的瘦素有可能通过增强受体的活性而增加下丘脑的转运及其信号传导,从而抑制NPY的表达。
Objective To investigate the expressions of Leptin, Leptin receptor(Ob-Ra, Ob-Rb), and neuropeptide Y(NPY)in the fat and hypothalamus of rats with chronic renal failure(CRF). Methods Forty Wistar rats were randomly divided into two groups: the normal control group(n = 15)and CRF group(n = 25). Fat and encephalic tissues were obtained to detect the gene and protein expressions of leptin, leptin receptor(Ob-Ra, Ob-Rb), and neuropeptide Y(NPY)with RT-PCR and immunohistochemistry. Results Compared with the control group, leptin gene expression significantly decreased in CRF group, while the protein expression significantly increased in fat tissue. Expressions of Ob-Ra and Ob-Rb in hypothalamus and coroid gland significantly increased while NPY decreased in CRF group. ConclusionsRemarkable metabolic disorders of NPY, leptin, and leptin receptors exist in CRF rats. The excessive leptin may inhibit NPY expression by enhancing the activity of receptors and thus improving the transfer of hypothalamus and its capability of signal conduction.
