斑点免疫结合试验对蓝氏贾第鞭毛虫感染者粪中特异性抗原的检测 被引量:3

摘要 应用蔗糖密度梯度离心技术从蓝氏贾第鞭毛虫感染者的粪便中分离纯化贾第虫包囊,以此包囊免疫家兔,获得免疫血清,经大肠杆茵吸收后建立斑点免疫结合试验,检测粪中贾第虫特异性抗原。以此法检测39例贾第虫感染者粪便,有36例出现阳性斑点,阳性检出率为92.3%。以超声粉碎的贾第虫包囊抗原为标准,36例阳性标本中,有35例标本贾第虫特异性抗原蛋白含量分布在39~69ng/mL范围内,1例为138ng/mL。 A dot immunobinding assay for detection of Giardia lamblia specific anti- gen(GSA)in fecal eluates was developed. The rabbit antiserum to Giardia lamblia cystwhich had been absorbed with Escherichia coli was used in the test,The positive rate in microscopically confirmed cases was 92.3%(36/39).The estimated amount of antigen found in almost positive samples ranged from 39 ng to 69 ng Giardia protein per millil-itre ,except one with 138 ng per millilitre compared with results of serially diluted cystantigen.The amount of soluble cyst antigen as low as 3.9ng/mL could still be detected by this test.
