摘要
比较了应用改良Tyrode′s液(简称T,对照组)与0.6%BSA-TCM199(简称BT)作为受精液(实验1)、这两种受精液的不同配比作为受精液(T:BT分别为3:1、1:1和1:3;T为对照组,实验2)和在不同受精时间采用不同受精液(先用T,IVF24h后改用BT或TCM199+10%发情牛血清,实验3),对经体外成熟培养(IVM)后的牛卵母细胞体外受精(IVF)效果的影响。结果,应用成分复杂的BT作为受精液,可以提高受精卵体外囊胚发育率(55.2%,对照组为37.1%,P<0.01);但受精卵卵裂率(41.8%)却极显著地低于对照组(79.4%,P<0.001);当应用T:BT为3:1和1:1的混合液作为受精液时,受精卵仍保持较高的囊胚发育率(分别为53.1%和57.4%,对照组为42.2%);而受精卵卵裂率(分别为79.1%和67.0%)虽比对照组(84.5%)低,但明显高于单纯用BT组(41.8%),从而使总的囊胚率得到提高。而在IVF24h后用BT或TCM199+10%发情牛血清将原先受精用的T液更换,再继续进行20h或46h的受精培养,则对体外受精无任何改善作用。结果说明,在现行使用的、成分简?
The
experiments were separately conducted to determine the effects offertilization media which
included modified Tyrode's solution(T),BSA+TCM-199(BTCM)and T +BTCM mixtures in different
proportions(3:1,1:1 and 1:3,V/V)on the fertilization of bovine oocytes in vitro. The results
showed that the embryos at 2-8-cell stage fertilized in BTCM media had significantly high
capacity to develop to theblastocyte stage(55.8%)than those fertilized in modified Tyrode's
solution(37.1%),though the cleavage rate(CR) in BTCM group (41.8%)was much lower than that
in Tgroup(79.4%,P<0.001). When the oocytes were fertilized in T+BTCM mixtures in 3:1 or 1:1
proportion,not only were the development rate of blastocysts from 2-8-cellembryos kept at a
high level(53.7%and 57.4%respectively),but the oocytes'CR wasalso high(79.0%and
67.0%respectively). It showed that the quality of 2-8-cell em- bryos can be improved when the
bovine oocytes were fertlized in the medium containing0.6%BSA and 25%to 50%TCM-199, and
more blastocysts can be obtained in vitrc。
