摘要
目的研究血管紧张素Ⅱ(AngⅡ)对人心房细胞[Ca2+]i的影响以及AngⅡ受体拮抗剂大蒜素的拮抗作用。方法急性分离单个人心房肌细胞,实验分4组:对照组、AngⅡ组(加入终浓度为0.1μmol/L的AngⅡ)、大蒜素组(加入终浓度为50μmol/L的大蒜素)和AngⅡ+大蒜素组(大蒜素50μmol/L和AngⅡ0.1μmol同时加入)。以Fluo-3/AM荧光指示剂负载,应用激光共聚焦显微镜技术,分别于加入干预药物后即刻与15min检测[Ca2+]i变化。结果对照组和大蒜素组人心房肌细胞内荧光强度和光密度值较低。AngⅡ加入后即刻光密度值开始增加,15min后荧光强度和光密度值显著增高,差异有显著性意义(P<0.05)。而AngⅡ+大蒜素组光密度值低于AngⅡ组,差异有显著性意义(P<0.05)。结论AngⅡ可引起人心房肌细胞内钙超载,大蒜素能显著减轻AngⅡ诱导的人心房肌细胞内Ca2+超载。
Objective To explore effects of angiotensin (AngⅡ) on intracellular free calcium concentration([Ca2+]i) in human atrial myocyte and antagonism of garlic extract. Methods Human atrial myocytes were isolated and divided into four groups,i.e,control group,Ang II group, garlic extract group and Ang Ⅱ+ garlic extract group. Confocal microscope was used with Fluo-3/AM as an indicator to detect changes of [Ca2+]i immediately and 15 minutes after adding Ang II or/and garlic extract, respectively. Results Comparing with control group, intracellular fluorescent intensity of myocyte was no change in garlic extract group, and was significantly higher in AngⅡ group 15 minutes after adding Ang II (P<0.05). Value in Ang Ⅱ+ garlic extract group was significantly lower than that in AngⅡgroup (P<0.05). Conclusion Ang II may induce intracellular calcium overload in human atrial myocyte,which can be significantly reduced by garlic extract.
