晚期糖化终产物及其多克隆抗体的制备

摘要 目的制备晚期糖化终产物(advancedglycoslationendproducts,AGEs)及其多克隆抗体并鉴定其特异性用于糖尿病及其并发症的研究。方法牛血清白蛋白(BSA)与葡萄糖(100mg/L)孵育3个月和多点、多次免疫接种新西兰大耳白兔,所得抗血清用亲和层析纯化鉴定。结果AGEsBSA荧光值为5.26±0.21,而BSA为0.16±0.01。荧光光谱扫描显示AGEsBSA激发高峰位于365nm,发射高峰位于446nm。抗BAGEs抗体亲和层析前抗体滴度为1:128,亲和层析后抗体滴度为1:2(P<0.01)。结论所制备的晚期糖化终产物及其抗体效价高,质量稳定。为进一步研究糖尿病及其并发症提供了必要的物质基础。
