重组腺病毒Ad-huVEGF_(121)治疗兔股骨头坏死的实验研究

摘要 目的:探讨一种治疗股骨头缺血坏死的新方法。方法:重组腺病毒Ad-huVEGF121注射入坏死的股骨头,应用RT-PCR和免疫组织化学技术检测VEGF121的表达,用组织形态学技术观察坏死股骨头的修复过程。结果:转染后的股骨头内有VEGF121表达,组织形态学观察发现转染后血管生成明显增多,新骨形成明显多于对照组。结论:转染VEGF121基因可以增加骨组织内血管生成,坏死骨的修复速度增快。 Objective:To explore a new method for the therapy of vascular necrosis of the femoral head. Methods: The recombinant Ad-huVEGF 121 was implanted in the necrotic femoral head. The expression of vascular endothelial growth factor (huVEGF 121 ) was examined by RT-PCR and immunohistochemical techniques. Repair of the femoral head was observed by histomorphometric analysis. Results:The expression of huVEGF 121 was detected in the femoral head transfected with huVEGF 121 gene. A significant increase in angiogenesis and in new bone formation after gene transfection was observed. Conclusion:Transfection of VEGF 121 gene enhances bone tissue angiogenesis and accelerate the repair of osteonecrosis accordingly.
