摘要
目的观察大豆黄酮对人乳腺癌MCF-7细胞体外增殖及细胞周期的影响并探讨其作用机制。方法分别采用台盼蓝拒染法、流式细胞术,在不同条件下测定大豆黄酮对细胞增殖抑制率及细胞周期分布的影响。蛋白激酶C(PKC)活性通过测定转移到CK2底物上的[γ-32P]ATP的32P的放射性活度来检测。结果大豆黄酮能够显著地抑制MCF-7细胞的体外增殖,作用24h可使MCF-7细胞的细胞周期阻滞于G2/M期和S期。细胞内PKC活性分析表明:大豆黄酮能够显著地抑制MCF-7细胞内PKC的活性并呈剂量依赖性关系,其IC50为13.27μmol/L。结论大豆黄酮可抑制人乳腺癌MCF-7细胞的体外增殖,其作用机制可能与细胞周期G2/M期和S期阻滞及抑制PKC的活性有关。
Objective To observe the influence and the action mechanism of daidzein on proliferation and cell cycle of human breast cancer cells MCF-7.Methods In the various conditions,the inhibitory rate of cell growth was tested by trypan blue stain assay and the influence of daidzein on cell cycle was observed by flow cytometry, respectively. The protein kinase C (PKC) activity was assayed by detecting incorporation of ^(32)P of[γ-^(32)P]ATP into the substrate. Results Daidzein inhibited the proliferation of MCF-7 cells strongly. After being treated for 24 hours with daidzein, MCF-7 cell cycle progression were arrested at G_2/M and S phase, which was confirmed by flow cytometry. The analysis of PKC activity showed that daidzein was shown to strongly inhibit the endogenous activity of protein kinase C in a dose dependent manner with IC_(50) of 13.27 μmol/L. Conclusion Daidzein could inhibite the proliferation of human breast cancer MCF-7 cells in vitro, the role of daidzein may be the two pathways of cell cycle blocking and PKC activity inhibiting. 【
