摘要
目的探讨体外受精-胚胎移植中卵泡颗粒细胞凋亡与妊娠率的关系。方法随机选择体外受精-胚胎移植中60例患者,收集其颗粒细胞,采用流式细胞技术检测颗粒细胞亚二倍体的含量和PDCD5的荧光密度;颗粒细胞免疫组化检测凋亡相关蛋白Bcl-2、Bax、PDCD5的表达。比较妊娠组和非妊娠组年龄、基础FSH值、促性腺激素的用量、取卵总数、受精率、妊娠率和颗粒细胞亚二倍体细胞的百分率、颗粒细胞PDCD5荧光密度、颗粒细胞Bcl-2、Bax、PDCD5蛋白表达的积分光密度值。结果妊娠组和非妊娠组年龄、基础FSH值、促超排卵FSH的用量、取卵数、受精率及免疫组化Bax积分光密度比较,差异无显著性(P>0.05),颗粒细胞亚二倍体百分率、颗粒细胞PDCD5荧光密度、PDCD5蛋白表达的积分光密度值妊娠组低于非妊娠组,颗粒细胞Bcl-2表达的积分光密值妊娠组高于非妊娠组,差异有显著性(P<0.05)。结论体外受精-胚胎移植中卵泡中颗粒细胞凋亡与妊娠率有关,凋亡率高,妊娠率低。
Objective To explore the relationship between granulosa cells apoptosis and the pregnancy rate in in-vitro fertilization and embryo transfer (IVF-ET). Metho ds Sixty infertility cases receiving IVF-ET were used to collect data of granulosa cells. The experiment was carried out through the following procedu res: 1) tests of the subG1 content and PDCD5 monoclonal antibody fluorescence de nsity of the granulosa cells with flow cytometry; 2) tests of the expression of pro- and anti-apoptosis proteins PDCD5, Bcl-2, Bax in granulosa cells by usin g immunohistochemistry. Comparisons of age, basal serum FSH concentration, numbe r of ampules of Gn administered, number of retrieved oocytes, cumulative concept ion rate, fertilization rate, subG1 content, PDCD5 monoclonal antibody fluoresce nce density, and expression of PDCD5, Bcl-2 and Bax in granulosa cells were mad e between fertility and infertility groups. Results There was no significant difference between the two groups in terms of age, basa l serum FSH concentration, number of ampules of FSH administered, number of retr ieved oocytes, cumulative conception rate, fertilization rate and Bax protein ex pression in granulosa cells. However, the fertility group had lower subG1, PDCD5 monoclonal antibody fluorescence density and PDCD5 proteins expression in granu lose cells and higher Bcl-2 proteins expression in granulose cells compared to the fertility group. Conclusion Good relationship exists b etween the granulosa cell apoptosis and pregnancy rate in IVF-ET. Pregnancy rat e shows a decreasing trend with the increase in apoptosis of granulosa cells.
