碘油抗癌药化疗栓塞剂的理化特性研究 被引量:13

摘要 分别将40%碘油(LIP)及超液态碘油(UFL)与丝裂霉素C(MMC)用“泵法”配制成几种不同的乳剂与混悬剂。将各种制剂用肉眼与显微镜观察其物理稳定性,用溶出试验仪以桨叶法进行药物释放试验,释放的MMC浓度用分光光度法测定。结果表明,碘油抗癌药化疗栓塞剂的各种制剂中无乳化剂的乳剂最不稳定,油水易分层,药物释放很快;混悬剂其次;含乳化剂的乳剂稳定性最好。油水比对乳剂的稳定性有明显影响,4:l的乳剂比2:1者稳定;用UFL制成的乳剂优于用LIP制成者,特别是加入乳化剂后乳剂很稳定,2小时MMC的释放量仅10%左右,据此可增加其中的抗癌药剂量,使抗癌药长期维持一定浓度作用于肿瘤细胞,又不致于增加副作用。 Several types of emulsions and suspensions were prepared using 40%Lipiodol(LIP)Or Ultra-Fluid Lipiodol (UFL)and mitomycin C(MMC)by the“pumping technique”The physical stability of each preparation was examined macroscopically and microscopically.The drug release test was made using dissolution tester by rotating oar method,the concentrations of MMC released from the preparations were determined spectrophotometrically. The results showed that the emulsion without emulsifier was the most unstable, undergoing phase separation easily and releasing the drug too rapidly;coming next was the suspension. The emulsion with emulsifier was the most stable. The stability of the emulsion was closely related to oil-water ratio, the emulsion with oil一water rate of 4:l was stabler than that of2:1. The stability of the UFL emulsion was better than that of the LIP emulsion especially the UFL emulsion with emulsifier which released,only about l0%of MMC in 2 hours. This result sug-gested that the dose of anticancerous drug in this type of emulsion with emulsifier can be increased in or-der to improve the therapeutic effect without concurrent increase of adverse reactions.
