摘要 目的探讨肿瘤转移抑制基因KiSS1对卵巢癌细胞株HO8910生物学行为的影响。方法含有人KiSS1基因全长cDNA的重组真核表达载体pcDNA3KiSS-1经酶切和测序鉴定正确后,转染卵巢上皮癌细胞株HO8910,观察其KiSS1mRNA表达、细胞生长及增殖能力和细胞侵袭力的变化。结果pcDNA3KiSS1成功转染HO8910细胞,KiSS1mRNA表达阳性,而亲本HO8910细胞为阴性。转染目的基因后细胞生长曲线、软琼脂克隆形成率等与未转染之细胞无显著性差异(P>0.05),但细胞侵袭能力明显下降(P<0.01),穿透Matrigel膜细胞数较未转染组明显减少(P<0.01)。结论KiSS1基因对卵巢上皮癌细胞HO8910的生长、增殖能力无影响,但可抑制其侵袭能力。 Objective To investigate effect of KiSS-1 gene on the biological behaviors of ovarian cancer cell HO8910.Methods The recombinant eukaryotic expression vector(pcDNA3-KiSS-1) inserted by the full-length cDNA encoding human KiSS-1 gene was constructed and confirmed by enzyme digest identification and DNA sequencing. The pcDNA3-KiSS-1 vector was transfected into human ovarian cancer cell line HO8910 by lipofectamine. The characteristics in the KiSS-1 transfected cells including expression of KiSS-1 mRNA, ability of growth, proliferation and invasion were observed. Results KiSS-1 gene was integrated into the genomic DNA of HO8910. Expression of KiSS-1 mRNA was positive in transfected cells and negative in untransfected cells. In comparison of cell proliferation curves and clone formation rate between the transfected and untransfected group there was no significant different found. Boyden Chamber invasion assay showed that cell number invading through Matrigel filter was significantly decreased in the KiSS-1 transfected group compared with that in the untransfected group. Conclusion KiSS-1 gene suppresses invasion of ovarian cancer cell HO8910 in vitro without affecting growth and proliferation.
