摘要
目的 研究慢性氟中毒对神经细胞损伤的作用机理及与细胞凋亡的关系。方法 利用RT PCR方法观察细胞凋亡相关基因B细胞淋巴瘤 /白血病 x(Bcl x)和白细胞介素 1β转换酶 (ICE)在慢性氟中毒鼠脑组织中的表达水平。结果 慢性氟中毒时脑组织中的Bcl x表达未受明显影响 ;ICE则明显高于正常对照组。结论 慢性氟中毒所致的神经细胞凋亡可能与激活G蛋白或氧化应激有关。当发生慢性氟中毒时 ,无明显证据表明Bcl x参与了此过程 ,但也不除外Bcl x在参与慢性氟中毒对神经细胞的损害过程中发挥作用并调节其表达水平达到平衡的可能性 ;ICE则可能参与了神经细胞发生凋亡的过程 。
Objective To study the action mechanism of chronic fluorosis in neurones and infection factors of apoptosis.Methods Method of molecule biology:using of reverse transcription polymerase chain reaction(RT PCR)observed expression level of correlation gene in apoptosis; they are Bcl x and interteukin 1β convert enzyme(ICE).Results The correlation gene of apoptosis determine results indicate: expression level of Bcl x of chronic fluorosis group in brain does not suffer to effect obviously; expression level of ICE of chronic fluorosis group in brain is higher than control group distinctly.Conclusions The mechanism of neurone apoptosis from the chronic fluorosis is likely relative with active G proteins and oxidative stress. When the brain appeared chronic fluorosis, no evidence indicated that Bcl x enlisted the process. But there is a possibility when Bcl x is concerned with the process of neurone damage from chronic fluorosis and exerted function, Bcl x regulate its express level to normal at the same time. ICE participated in the process of neurone apoptosis from chronic fluorosis and bring into play the main action.
