锰超氧化物歧化酶基因V(16)A多态性的种族差异及与糖尿病肾病的关系 被引量:3

摘要 目的:探讨锰超氧化物歧化酶(MnSOD)基因V(16)A多态性的种族差异及与糖尿病肾病(DN)的关系.方法:在中国昆明地区汉族人中对187例2型糖尿病患者(其中正常白蛋白尿即DN0组50例,微量白蛋白尿DN1组64例,大量白蛋白尿即DN2组73例)和97例健康对照者的MnSOD基因V(16)A多态性进行检测,并比较不同种族中该位点基因型和等位基因分布及频率.结果:(1)中国昆明地区汉族人中存在MnSOD基因V(16)A多态性;(2)与日本人群、俄罗斯人群相比其基因型和等位基因频都存在显著性差异,中国和日本人群以VV型为主,俄罗斯人群以AA和AV型为主;(3)DN组(DN1+DN2)VV基因型和V等位基因频率显著高于不伴肾病的DN0组(分别为χ2=6·42和5·08,P<0·05),Logistic回归分析表明VV基因型相对于VA+AA基因型是DN发生的遗传危险因素.结论:MnSODV(16)A多态性分布存在种族异质性,在中国昆明汉族人2型糖尿病患者中MnSOD基因V(16)A多态性与DN发生相关.不同种族人群MnSODV(16)A多态性分布的差异,可能是亚裔人群比白种人群2型糖尿病患者DN发病率高的原因之一. Objective:To investigate the ethnic variation in V16A polymorphism of manganese superoxide dismutase(MnSOD) gene and theassociation between this polymorphism with diabetic nephropathy(DN).Methods:V16A genotype of MnSOD was assayed in 187 type 2 diabetic patients(including 50 normal albuminuria in DN0 group, 64 microalbuminuria in DN1 group,73 macroalbuminuria in DN2 group) and 97 healthy nondiabetic subjects in KunMing Han Chinese. The distribution frequencies of the polymorphism were compared with different racial. Results:(1) There was V16A polymorphism of MnSOD gene in KunMing Han Chinese. (2) There were significant variations in this polymorphism among three different ethnic groups. The VV type and V allele frequency was significantly higher in Asian (Chinese and Japanese) than in European(Russian).(3)The 16VV genotype and 16V allele frequencies in the diabetic patients with nephropathy were significantly higher than without nephropathy(χ~2=6.42, 5.08,P<0.05, respectively).Furthermore, logistic regression analysis showed that this polymorphism was significantly correlated to DN. Conclusion The were ethnic variations in V16A polymorphism of MnSOD gene.The MnSOD gene V16A polymorphism may be associated with the occurrence of diabetic nephropathy in Kunming Han Chinese. The variation frequency of MnSOD gene V16A polymorphism among different ethnic groups may be one of the reasons in the racial different incidence of diabetic nephropathy.
