摘要
目的 研究脑出血时脑组织生长抑素mRNA的表达水平,探讨电针治疗脑出血后脑损伤的可能作用机制。方法在复制易卒中型高血压模型的基础上,以胶原酶加肝素脑内注射诱发脑出血,建立高血压性脑出血大鼠模型。运用逆转录聚合酶链反应技术检测电针治疗后不同时相点脑出血大鼠海马生长抑素mRNA的表达,并以正常组、假手术组和模型组加以对照。结果 高血压性脑出血模型组大鼠海马生长抑素mRNA的表达明显减弱,电针治疗后其表达显著增强,与模型组同时相比较有显著意义,并随着时间的推移而逐渐趋于恢复。结论 电针水沟、内关、足三里穴可以通过增强脑出血脑组织生长抑素基因转录水平,增加生长抑素合成,调节神经元活动和神经营养而改善脑出血后脑组织损伤程度。
Objective To study the somatostatin mRNA expression in the brain tissue of cerebral hemorrhage and the mechanisms of acupuncture on the brain injury following cerebral hemorrhage.Methods The hypertensive cerebral hemorrhage rat model was evoked by injecting collagenase and heparin into the brain of stroke prone hypertensive model rat. The expression of somatostatin mRNA in the hippocampus of the rats with hypertensive cerebral hemorrhage after acupuncture treatment was detected with reverse transcription polymerase chain reaction on different phases and compared with those of the normal group, sham operation group and model group.Results The expression of somatostatin mRNA in the hippocampus of the rats with hypertensive cerebral hemorrhage was significantly weaker than those of normal and sham operation group, while after electroacupuncture treatment, it was significantly increased with the lapse of time.Conclusion Electroacupuncture with Shuigou(GV 26), Neiguan(PC 6) and Zusanli(ST 36) acupoints may improve the brain tissue damage by promoting the gene transcription of somatostatin and increasing the synthesis of somatostatin to improve the activities and nutrition of the neurons.
