摘要
目的探讨参麦注射液对培养鼠大脑皮层神经细胞缺氧损伤的保护作用。方法将培养8d后生长良好的神经细胞随机分为正常组、模型组、参麦组和尼莫地平组;每组设3个培养瓶,正常组不作任何处理,其余3组均分别在缺氧后30min、1h、3h、6h和12h5个时间点造成神经细胞缺氧损伤模型。参麦注射液(1∶50)和尼莫地平(20μg/mL),分别在缺氧前24h及缺氧后加入,相差显微镜下观察神经细胞形态学的变化,测定DNA裂解率。结果参麦组和尼莫地平组的神经细胞在缺氧损伤后的形态学改变明显比模型组轻,DNA裂解率与模型组相比亦有显著性差异(P<0.05)。结论参麦注射液可能通过抑制细胞凋亡而对缺氧损伤的鼠大脑皮层神经细胞具有保护作用。
Objective To investigate the effect of Shenmai injection on hypoxic injured cortical neurons in cultured rats.Methods The neurons cultured for 8days in better condition were randomly divided into 4 groups:normal group,modeling group, treating group with Shenmai injection and treating group with Nimodipine with 3 bottles in each group;except of normal group, the others were made into hypoxic models respectively in 30min、1h、3h、6h、12h after hypoxia; Shenmai Injection(1∶50) and Nimodipine (20μg/mL) were added into culturing fluid of neurons 24h before hypoxia and after hypoia immediately;the morphological changes of neurons were observed by inverted/phase contrast microscope and the DNA splitting rate was determined.Results The morphological changes of Shenmai group and Nimodipine group were not so evident as those of modeling group;there was obvious significant difference.Conclusions Shenmai injection holds a protective effect on the hypoxic injured cortical neurons in cultured rats,which may be achievd by inhibting apoptosis.
