一株来自深海沉积物的低温、嗜压菌的分离鉴定 被引量:3

摘要 从西太平洋5060m深的海底沉积物中分离到一组低温菌,对其中一株细菌WP02225进行了生理生化和分子水平鉴定.结果表明:该菌革兰氏染色阴性;温度生长范围在28℃以下,最适生长温度15℃,pH耐受范围为4.5~9.5;最适pH值7.5.能够利用D葡萄糖、甘油和麦芽糖作为唯一碳源生长并产酸,氧化酶和接触酶呈阳性,可以水解明胶和还原硝酸盐,但不能继续还原亚硝酸盐,产H2S气体.依据其生理生化性质、系统发育学、16SrDNA序列同源性分析初步鉴定该菌属于深海中特有的菌属:Moritella属,与嗜压菌Moritellajaponica种有99.47%的同源性.生理生化指标表明,这两株菌不完全相同,Moritellajaponica可以利用果糖、不能利用麦芽糖,而WP02225正好相反.另外,两者的最适生长温度也不一致,前者属于嗜冷菌的范畴;后者属于耐冷菌. A kind of psychrophilic bacteria,Stain WP02-2-25,has been isolated from western Pacific Ocean sediment at a depth of 5 060 m,and identified by its molecular and phenotypic characteristics.The results showed that its growth temperature was under 28℃,and the optimal temperature was determined to be at 15℃,the optimal pH at 7.5.It belongs to be Gram negative,with positive affect by catalase and cytochrome oxidase.This kind bacterium can utilize D-glucose,glycerol and maltose as sole or principal carbon sources and generate acid at the same time.It can hydrolyze gelatin and reduce nitrate to nitrite.But,it can’t use nitrite to produce H_2S during growth.According to the phenotypic and phylogenetic analysis,this stain belongs to the genus Moritella,and closest to a barophilic species,M.japonica.However,they are different in utilizating carbohydrates.Our stain is unable to utilize fructose but able to use maltose,while M.japonica is just switched.In respect of its optimal temperature over 10℃ the isolate was classified to be psychrotrophs.
