摘要
以茎段、茎尖为外植体,在WPM培养基中加入不同浓度的玉米素(ZT),对越桔(Vaccinium vitis-idaea)进行了组织培养快速繁殖技术的研究.结果表明:以WPM+ZT 2 mg·L-1为越桔的适宜增殖培养基,增殖率为4~5倍;壮苗培养基为不加激素WPM培养基.在诱导生根时,以1/2 WPM+IBA 0.6 mg·L-1为最适宜的培养基,生根率为92%.以松针土+苔藓体积比1:1为基质时,组培苗移栽成活率最高,达到90%.
The technology of rapid propagation of Northland Vaccinium vitis-idaea was studied by tissue culture. The culture medium is WPM supplemented with different concentrations of ZT respectively or commixture. Explants were new buds, tips of stem and sections of stem respectively. The best medium for Northland propagation is WPM+ZT (2.0 mg·L^(-1)). And propagation rate is 4~5 times. Adventitious buds are cut as individual plant and planted in WPM medium without hormone before producing root. Adventitious root was produced in 1/2 WPM+IBA (0.6 mg·L^(-1)). Rooting rate is 92%. Survival rate of tube-seedling is the highest and reached 90%, transplanting in moos and pine needle soil(1∶1).
