怀化侗族高血压高发人群G蛋白β3亚单位825C/T多态性与血脂水平的关系 被引量:13

Association between G-protein beta3 subunit 825C/T polymorphism and blood lipids among the high risk hypertensive population of Dong nationality in Huaihua area

摘要 目的:探讨湖南怀化侗族高血压高发人群原发性高血压与G蛋白β3亚单位825C/T多态性与血脂水平以及频率分布和地域及人种差异的关系。方法:实验于2003-06/2004-12在怀化医学高等专科学校分子生物研究室进行。高血压高发人群为湖南怀化地区通道侗族自治县1个独立自然村落30岁以上人群,均为侗族,共185人,其中原发性高血压96人,血压正常者89人;另外选择怀化地区汉族30岁以上的227人作为对照组,其中原发性高血压76人,血压正常者151人。采用聚合酶链反应结合限制性内切酶片段长度多态分析方法,检测被试者的G蛋白β3亚单位825C/T等位基因频率和基因型频率,同时测定血脂和相关的生化指标,并进行病例对照分析。结果:按意向处理分析,怀化侗族人群185人和汉族人群227人的资料全部进入结果分析。①G蛋白β3亚单位825C/T基因型频率和等位基因频率:怀化侗族人群中原发性高血压患者和血压正常人的基因型频率比较无差异(CC:0.188,0.247,CT:0.594,0.528,TT:0.218,0.225,P>0.05),等位基因频率比较也无差异(C:0.484,0.511;T:0.516,0.489,P>0.05);汉族人群中原发性高血压患者和血压正常人的基因型频率比较无差异(CC:0.211,0.245,CT:0.605,0.642,TT:0.184,0.113,P>0.05),等位基因频率比较也无差异(C:0.513,0.556;T:0.487,0.434,P>0.05)。②G蛋白β3亚单位基因825C/T多态与各生化指标的关系:怀化侗族高血压高发人群G蛋白β3亚单位825C/T基因型与血糖、血脂和电解质水平无相关性(P>0.05),但是侗族原发性高血压患者与血压正常者比较低密度脂蛋白、总胆固醇和血钠偏高而血钾偏低(P<0.05)。③G蛋白β3亚单位825C/T基因型与不同地域人群的比较:其等位基因频率与中国北京人、东北人及日本人群接近,显著高于白种人(P<0.01)结论:怀化地区侗族与汉族人群原发性高血压和正常人之间G蛋白β3亚单位基因825C/T基因频率和基因型频率均无明显差异,且与日本和中国北方人群接近,而与白种人群有较大差异。怀化地区侗族原发性高血压患者低密度脂蛋白、总胆固醇和血钠偏高而血钾偏低,但是这些生化指标的异常与825C/T基因型无关,而该人群较高的原发性高血压患病率可能与血清低密度脂蛋白、总胆固醇、血钠增高、血钾降低以及钠/钾比值增高有关。 AIM:To investigate the relationship of G-protein beta3(β3) subunit 825C/T polymorphism and essential hypertension with blood lipid level,frequency distribution,differences of area and race in the high risk hypertensive population of Dong nationality in Huaihua area of Hunan province.METHODS:The study was carried out in the research room of molecular biology of Huaihua Medical College from June 2003 to December 2004.The high risk population of hypertension were 185 people of Dong nationality above 30 years old,including 96 cases of essential hypertension and 89 cases of normal blood pressure,who were the residents of an independent natural village in Tongdao Dong autonomous county of Huaihua area,Hunan Province.Another 227 Han subjects above 30 years old in Huaihua area were taken as controls(control group),including 76 cases of essential hypertension and 151 cases of normal blood pressure.The G protein β 3 subunit 825C/T allele frequency and genotypic frequency of the subjects were detected with reverse transcription polymerase chain reaction combined with restriction fragment length polymorphism analysis,blood lipid and related biochemical indexes were also determined,and the case control analysis was undertaken.RESULTS:According to intention to treat analysis,the data of 185 Dong subjects and 227 Han subjects in Huaihua area were all involved in the analysis of results.① The G protein β 3 subunit 825C/T allele frequency and genotypic frequency:For the subjects of Dong nationality in Huaihua area,there were no significant differences in the genotypic frequency(CC:0.188,0.247;CT:0.594,0.528;TT:0.218,0.225;P >0.05) and allele frequency(C:0.484,0.511;T:0.516,0.489;P >0.05) between patients with essential hypertension and subjects with normal blood pressure;Among the Han subjects,there were insignificant differences in the genotypic frequency(CC:0.211,0.245;CT:0.605,0.642;TT:0.184,0.113;P >0.05) and allele frequency(C:0.513,0.556;T:0.487,0.434;P >0.05) between patients with essential hypertension and subjects with normal blood pressure.② Association between G protein β 3 subunit 825C/T polymorphism and the biochemical indexes: Among the high risk population of hypertension of Dong nationality in Huaihua area, the G protein β 3 subunit 825C/T genotype had no correlation with blood glucose,blood lipid and electrolure levels(P >0.05);Compared with people with normal blood pressure,patients with essential hypertension of Dong nationality had higher low density lipoprotein cholesterol,total cholesterol and blood natrium but lower blood kaiu(P< 0.05).③ In comparison of G protein β 3 subunit 825C/T genotype between different area:Their allele frequency was close to that of people living in northern and northeast China and that of Japanese people, but significantly higher than that of white people(P< 0.01).CONCLUSION:The G protein β 3 subunit 825C/T allele frequency and genotypic frequency between patients with essential hypertension and subjects of normal blood pressure are not obviously different in both Dong and Han peoples in Huaihua area,and close to those of Chinese people living in the north and Japanese people,but greatly different from those of white people.Patients with essential hypertension of Dong nationality in Huaihua area have higher low density lipoprotein cholesterol,total cholesterol and blood natrium but lower blood kaiu,but the abnormalities in these biochemical indexes are not correlated with the 825C/T genotype;The higher prevalence rate of essential hypertension in these people may be associated with the increased low density lipoprotein cholesterol,total cholesterol and blood natrium,decreased blood kaiu,and the increased ratio of natrium to kaiu.
