摘要
目的观察大鼠视网膜缺血再灌注(retinalischemiareperfusion,RIR)后,凋亡即早基因cfos、cjun及白介素1β(IL1β)多肽在视网膜各层细胞中的表达变化,进一步明确凋亡即早基因和白介素1β与RIR损伤发生机制的关系。方法采用前房灌注生理盐水,形成130mmHg(17.3kPa)高眼压,诱导大鼠视网膜缺血60min,解除高眼压,建立RIR模型。缺血60min,再灌注1h、3h、6h、12h、24h后作视网膜石蜡切片,观察cfos、cjun及IL1β的免疫组化结果。结果正常对照组未见cfos、cjun及IL1β的表达,RIR后,cfos、cjun在视网膜神经节细胞层(ganglioncelllayer,GCL)和内核层(innernuclearlayer,INL)的表达为,再灌注1h少量表达,3h达到高峰,6h开始下降,12h、24h几乎趋于正常组表达。IL1β在再灌注12h、24h出现表达。而外核层(outernuclearlayer,ONL)几乎均无无阳性表达。结论cfos、cjun及IL1β参与RIR损伤的发生,RIR中视网膜节细胞层和内核层细胞存在凋亡征象。
Objective To determine in rats whether there is a change in c-fos and c-jun immediate early gene and interleukin-1β expression in retina after retinal ischemia reperfusion.Methods Retinal ischemia was induced in SD rat by means of raising the intraocular pressure to 130 mmHg(17.3 kPa) for 60-minute. Via cannulation into the anterior chamber, the model of retina ischemia reperfusion was established once releasing the pressure in the rat. The expression of c-fos, c-jun and IL-1β in paraffin section of retina was observed at 1 h, 3 h, 6 h, 12 h, 24 h after 60-minute ischemia by immunohistochemistry method.Results The expression of c-fos and c-jun first appeared in ganglion cell layer and inner nuclear layer in 1 h group, but not in outer nuclear layer. In 3 h group the expression of c-fos and c-jun arrived to the peak, and then decreased gradually in 6 h group. In 12 h and 24 h group, the expression of c-fos and c-jun arrived to normal level. But IL-1β appeared in ganglion cell layer and inner nuclear layer in 12 h and 24 h group. Weak signals for c-fos, c-jun and IL-1β were observed in the normal group.Conclusion It suggests that changes in expression of c-fos, c-jun and IL-β after retinal ischemia reperfusion,the phenomenon apoptosis is obvious in ganglion cell layer and inner nuclear layer after retinal ischemia reperfusion injury.
