摘要
目的探讨同时参与肺腺癌不同癌变进程的基因群,以期进一步筛选出肺腺癌的分子靶标。方法选取术后临床分期为Ⅰ、Ⅱ、Ⅲ期的肺腺癌组织标本共10例为实验组,相应的癌旁组织10例为对照组。与含13824个基因的基因芯片进行杂交,最后结合分期和临床预后对海量数据进行数据挖掘。在不同分期及不同预后的标本中均差异表达的基因即为入选基因。结果10例肺腺癌标本共差异表达基因119条,其中,含已报道与肺癌相关的基因26条,尚未报道与肺癌相关的基因46条,完全未知功能的新基因47条。结论119条基因是肺腺癌发生、发展过程中的重要分子标记,是肺腺癌分子靶的候选基因。
Objective To study the genes involved in the whole process of carcingenesis and development of lung adenocarcinoma for biomarkers identification. Methods 10 lung adenocarcinoma samples,including pathologic stage Ⅰ ,Ⅱ,Ⅲ disease,were tested and their surrounding tissue were also tested as control. Total RNA was extracted and labeled with fluorescence during reverse transcription. After hybridized on 20 slides of microarray with 13824 genes, the cy3:cy5 ratios were obtained by computer analysis. Each experiment was repeated two times and the mean value was chosen. Based on the data ,we analysed the expression profiles combined with qathologic stage and clinical prognosis. The differentially coexpressed genes in samples at different stages and with different prognosis were the target genes. Results 119 genes were identified which involved in many cell activities,such as growth regulation, signal transduction, metastasis, immunity, metabolism and so on. Among these target genes, the expression of 22 genes were upregulated,including 9 reported lung cancer related genes, 12 unreported genes and 1 unknown gene;the expression of 97 genes were downregulated,including 17 reported lung cancer related genes,34 unreported genes and 46 unknown genes. Conclusion The 119 genes play an important role in the whole process of carcinogenesis and development of lung cancer and can be used as candidate biomarkers for lung cancer.
