清肺口服液对3I、7b型腺病毒感染人胚肺成纤维细胞TGF-β_1mRNA基因表达的影响 被引量:2

摘要 目的:采用治疗小儿病毒性肺炎痰热闭肺证的有效中药制剂清肺口服液,研究其对腺病毒3I、7b感染的人胚肺成纤维细胞的转化生长因子-β1(TGF-β1)mRNA基因表达的影响。方法: 以3I、7b型腺病毒分别攻击体外培养的人胚肺成纤维细胞,用清肺口服液含药血清作用于该细胞,另设正常细胞组、病毒对照组、利巴韦林组,采用原位杂交法检测不同组细胞TGF-β1的mRNA 基因表达,并进行比较。结果:病毒对照组较正常细胞组的TGF-β1mRNA基因表达明显增强(P <0.01),含药血清组可显著降低3I、7b型腺病毒攻击后细胞TGF-β1的mRNA表达(P<0.01)。结论:腺病毒感染可使人胚肺成纤维细胞TGF-β1mRNA表达增高;清肺口服液可下调TGF-β1的mR- NA表达,这可能是其抗病毒作用的机制之一。 Objective: To study the effect of Qingfeikouhuye (Clearing - lung Oral Liquid), an efficacious TCM preparation for the treatment of the disease of child' s phlegm - heat obstructing lungs of viral pneumonia, on the gene expression of the transforming growth factor - β1 (TGF - β1) mRNA of the fibroblast cells of human embryonic lungs induced by aden-ovirus Types 3I and 7b. Method: Adenovirus Types 3I and 7b are respectively used to attack the fibroblast cells of human embryonic lungs, which are artificially cultivated in vitro and the medicine - contained serum of Qingfeikouhuye is added to the attacked cells and then the TGF - β1 mRNA gene expression of cells in such experimental groups as the group of normal cells, the control group of virus and the group of Ribavirin is detected by the method of in situ hybridization and compared among them. Result: The TGF - β1 mRNA gene expression of the control group of virus is tangibly higher (p <0. 01) than that of the group of normal cells, and TGF - β1 mRNA of the cells attacked by aden-ovirus types 3I and 7b of the group of the medicine - contained serum of the liquid apparently lowers down (p <0. 01) . Conclusion: TGF - β1 mRNA gene expression of the fibroblast cells of human embryonic lungs induced by adenovirus may heighten while Qingfeikouhuye is able to make the TGF - β1 mRNA gene expression come down. And this may be one of the liquid's mechanisms resistant to virus.
