摘要
目的检测缺氧诱导因子_1α(HIF_1α)及VEGF、CD34在乳腺癌的表达,探讨其与缺氧情况下乳腺癌血管生成及其与生物学行为的关系。方法采用免疫组织化学S_P法检测68例乳腺癌组织中HIF_1α及VEGF的表达,并用CD34抗体标记血管内皮细胞,计数乳腺癌组织中MVD。结果68例乳腺癌标本中有37例(54%)表达HIF_1α;HIF_1α表达组VEGF及CD34表达强度均高于HIF_1α阴性表达组;HIF_1α表达与肿瘤的分化程度及淋巴结转移密切相关(P<0.01)。HIF_1α表达与VEGF、CD34表达之间存在显著相关性。结论HIF_1α在乳腺癌组织中存在着过表达,部分的参与了乳腺癌的演进;HIF_1α通过调节VEGF的表达,在乳腺癌血管生成中发挥重要作用;HIF_1α、VEGF的过表达及高微血管密度(MVD)与乳腺癌的不良发展有关,可能成为预测乳腺癌侵袭、转移及预后的重要指标。
Objective To detect the expression of hypoxia-inducible factor-1α,VEGF and CD34 in breast cancer and discuss the relation between the angiogenesis in breast cancer and in biological action in case of anoxia.Methods We examined sixty_eight breast cancer specimen by adopting S_P immunohistochemical.We mark the cell in endothelium of blood vessel with CD34 antibody and count the MVD in the tissue of breast cancer.Results Thirty_seven cases in the sixty_eight breast cancer specimen express HIF_1α,The expression intensity of VEGF and CD34 in the positive expression group is higher than the negative expression group.It is closely related to the differentiation degree of tumor and metastasis of lymph node ( P <0 01).The expression of HIF_1α has a direct bearing on the expression of VEGF and CD34.Conclusions HIF_1α in breast cancer's tissue exists overexpression. It partially participates in the evolvement and development of breast cancer. HIF_1α plays an important role in the building of breast cancer's blood vessel by regulating the expression of VEGF.The overexpressions of HIF-1α and VEGF and MVD have relation with the ill development of breast cancer. It can be an important index for forecasting breast cancer's invasion, metastasis and prognosis.
