摘要
目的观察术后抗再狭窄药物紫杉醇对平滑肌细胞凋亡的影响及量效、时效关系。方法培养大鼠主动脉平滑肌细胞,培养至4到9代。根据紫杉醇作用时间不同将细胞分成24小时组和72小时组,每一时间组根据加入不同浓度紫杉醇分四组:0μmol/L,0.1μmol/L,1μmol/L,10μmol/L组。用流式细胞仪检测紫杉醇是否引起细胞凋亡,并观察此作用与药物浓度和时间的关系。结果紫杉醇作用VSMC24小时后0μmol/L组及低、中、高浓度紫杉醇组细胞凋亡率分别为2.2%、3.6%、6.8%、22.6%;72小时后细胞凋亡率分别为6.1%、10.4%、15.4%、42.7%。结论紫杉醇能促进平滑肌细胞的凋亡,随着药物浓度提高,作用时间延长,细胞凋亡率也相应升高。故促进平滑肌细胞调亡可能是紫杉醇抑制平滑肌细胞过度增殖的重要机制。
Objective Percutaneous transluminal coronary angioplasty (PTCA) is limited by restenosis. The effect of paclitaxel, which can prevent restenosis,on the VSMC apoptosis ,and the relation between result ,time and dosage were observed. Methods The VSMC were devided into two groups, the 24 hours group and 72 hours group.And there are four different dosage groups in every time group:0 μmol/L, 0.1 μmol/L,1 μmol/L and 10 μmol/L.The flow cytometer was used to observe the relation between result, time and dosage. Result That experiment has demonstrated that :the apoptosis rate of 0 μmol/L group, low, medium and high concentration paclitaxel group were 2.2%,3.6%,6.8% and 22.6% after 24 hours,6.1%,10.4%,15.4% and 42.7% after 72 hours. Conclusion Paclitaxel can induce cell apoptosis, and the higher dosage the longer the time,the higher apoptosis ratio accordingly. Accelerating cell apoptosis is the principal mechanism of paclitaxel preventing the SMC over proliferation.
