摘要
目的观察土贝母皂甙对乙型肝炎病毒(HBV)基因转染的人肝癌细胞系2215细胞的毒性及其分泌的HBsAg和HBeAg的抑制作用。方法以MTT比色法观察药物的细胞毒性,以酶联免疫吸附(ELISA)法检测培养上清中HBsAg和HBeAg的表达。结果不同浓度的土贝母皂甙对HBsAg和HBeAg的分泌均有一定的抑制作用,药物作用到48h时对HBsAg和HBeAg的抑制作用最强(P<0.01),且对HBeAg的抑制作用强于拉米夫定;在0.01ug/ml浓度下无明显细胞毒性作用。结论体外细胞培养表明,土贝母皂甙对HBV有抑制作用。
Objective To investigate the suppressor effect of tubeimoside on HBsAg and HBeAg in the cultured hepatocarcinoma cell line 2.2.15 which was gene transferred with hepatitis B virus (HBV) and its toxicity.Methods To observe the toxicity of medicine by using MTT chromometry and the expression of HBsAg and HBeAg in the cultivation supernatant by using Enzyme linked immunosorbent assay (ELISA).Results Tubeimoside had the suppressor effect on the 2.2.15 cell secreting HBsAg and HBeAg . At 48h, the suppressor effect is the strongest on HBsAg and HBeAg (P<0.01). At 48h and 72h, the suppressor effect is stronger than 3TC on the 2.2.15 cell secreting HBeAg; it is unobvious the effect of the cell toxicity under 0.01ug/ml.Conclusions The cell culture indicated that tubeimoside can inhibit hepatitis B virus (HBV) in vitro.
